At your 7th point, you are supposed to mention that complementary sticky ends are produced.
At your 10th point, the enzyme mentioned is DNA ligase.
At your 12th point, your have to mention that you need to apply temporal heat or shock to open up the pores of the bacteria's cell membrane and hence allow the recombinant DNA to enter the bacterial cell.
And in any case, a fermenters' actual process no need to know in depth just know that what are the conditions that the fermenter needs to keep in order to maintain an optimum conditions for the transgenic bacteria to grow and multiply quicky...i.e. ph, oxygen, temperature, nutrient broth that contains a carbon-containing energy source(glucose), nitrogen source(amino acids) and also mineral salts.
Bacterias are not used as vectors...rather the term vector is used to refer to another form of DNA that can be transferred from one organism to another, and in this case the vector is the plasmid isolated from the bacteria.
lol this topic coming out for essay issit?

pro to the max!
Don't talk cock, fermenter is old syllabus.
EDIT: Sorry, I talk cock. Win liao. My whole class sure fail if fermenters come out. Wait, it's my whole school lol. Never cover.
SEAB says:
5094/02: Learning Outcome: outline the process of large-scale production of insulin using fermenters.
At your 7th point, you are supposed to mention that complementary sticky ends are produced. YES.
At your 10th point, the enzyme mentioned is DNA ligase. You don't have to mention it.

To add on, mention:
insulin produced in bacterial cell --> extract --> purified --> for use

not your whole school.... now you know liao... win liao
well...its always better that you know more, you never know what may come out...like physics....
I thought DNA Ligase have to know and mention de?

My teacher just asked us to use the word 'enzymes'.
Just in case they ask for the production of something else with which a bacterial plasmid is used as a vector, you can use the common word: enzyme to prevent mistakes. It's safer, I think.

Ah, I'll keep my fingers crossed and hope for fermenter to come out.
Do us a favour, keep the secret, will you?

Never harms to study more, right?

Production of insulin by bacteria using genetic engineering
• Isolation of the gene of interest
• The gene that produces insulin is identified on the human DNA.
• Using restriction enzymes, this gene is ‘cut’ from the DNA
• A fragment of the DNA with sticky ends is produced. This fragment includes the insulin gene.
• Isolation of plasmid DNA from bacterial cell
• The plasmid DNA is isolated from the bacterial cell.
• Then, using the same restriction enzyme, the plasmid DNA is cut to produce a linear strand of plasmid DNA with sticky ends.
• Insertion of the human DNA into the plasmid DNA
• The cut human DNA fragment with the gene of interest (from [1]) is mixed with the cut plasmid DNA (from [2]).
• Using enzymes to join the sticky ends, the human DNA is inserted into the plasmid DNA. ie. A recombinant DNA is produced.
• Introduce recombinant DNA back into the bacterial cell
• The bacterium is treated so that the plasmid can enter the bacterial cell.
• Thus the plasmid acts as a vector to carry the human gene into the bacterium.
• Once this is successful, the bacterium will multiply and all the bacteria produced will have the human insulin gene in them.
• Each bacterial cell will start to make insulin molecule.
Thanks Theguiltshow, what an exquisitely detailed piece of crucial information you have yielded. I shall proceed to cram all these newfound info. into my brain now.
I think that thing will come out. hopefully for essay. And if it does, you do have to include those detailed info, since they are in the tb anyways.
Lol if it comes out for essay question then GG to all lol. And the details u can put if u can rmb it then ba. I think just write enzymes can le ba... Or u can be an asshole and write "enzymes (eg. DNA Ligase)" I think lol.
I love Bio
But I returned all to teacher le

My textbook seems to disagree. It says, and I quote: Mix the plasmid with the DNA fragment containing the insulin gene. Add DNA ligase to join the insulin gene to the plasmid.
If you've noticed, there was no mention of mixing the 'gene of interest' with the plasmid and insulin DNA fragment.

I hate essay questions that make you recall what you're supposed to have memorized. It makes me sick.

I think I can emphatize.
Eh? But I think essay questions on Chpter 17 not bad leh? Dunno can go toilet check de. Paste stickers wif the names of the parts on your lower region. Then go toilet check check lor.

Pasting stickers is painful. When you pull it off, you'll do permenant damage to your pubic hair. I recommend folding the pages of interest into little pieces and then stuffing it into your abdomen. Much more practical, I'd say.
Plus, you will never be detected.
LOL I didnt say paste at pubic hair. I said paste on the respective parts lol. The olvl examiners slack slack de...

lol. Biological graffiti? My girlfriend would be most pleasantly surprised.
Tellingly, do you think we're off-topic? I was very sure about entering a bio-panic thread.
Lol I think we are off-topic also -.- The TS offline le... Aiya going to study more bio le lol. L8r chat chat too much I die during o lvl l8r lol
